Indications and usage     

Pentoxifylline is a peripheral vasodilator drug. Increases the elasticity of red blood cells, inhibits the aggregation of the red cells and platelets and decreases blood viscosity, promoting blood circulation.

Pentoxifylline is indicated in the treatment of disorders of peripheral circulation, such as:

- Blood flow abnormalities of extremities (leg pain when walking and resting),
- Night cramps in the calves, tingling and numbness in the legs and arms),
- Ulcers (sores) in the legs and gangrene.

Medizzine recommends consult your doctor if you are unsure (or you do not know) why you have been given pentoxifylline.

Other ingredients of available commercial presentations:

Each tablet contains 400 or 600 mg pentoxifylline. The other ingredients may vary from one to another brand and may include: Hypromellose, microcrystalline cellulose, crospovidone, colloidal silicic anhydride, magnesium stearate, macrogol 6000, titanium dioxide, talc, 30% polyacrylate, carnauba wax, lactose, triacetin, erythrosine and indigo-carmine aluminum lake.

A 15 ml vial contains 300 mg pentoxifylline (INN) . As excipients, sodium chloride and water for injection.

Important information     

This medicine in tablets contain lactose. If your doctor has told you have an intolerance to some sugars, contact himr before taking it.

Before taking the drug     

Do not use pentoxifylline

If you are allergic (hypersensitive) to pentoxifylline or any of the other ingredients of the medicinal product prescribed to you. If you think may be allergic, ask your doctor.

Allergy symptoms may include:

- Asthma attacks with shortness of breath, audible wheezing or rapid breathing.
- More or less sudden swelling of face, lips, tongue or elsewhere in the body. It is especially critical if it affects the vocal cords.
- Hives, itching, rash
- Anaphylactic shock (loss of consciousness, paleness, sweating, etc.).

Nor you should take this medicine:

If you have severe bleeding or a severe retinal bleeding caused by diseases such as diabetes and high blood pressure.
If you have recently had a myocardial infarction

Take special care with pentoxifylline

- If you have severe heart rhythm disturbances
- Myocardial infarction,
- Arterial hypotension,
- Severe liver disease
- Severe kidney disease
- Increased tendency to bleeding due to anticoagulant medication or coagulation disorders

Taking other medicines

Tell your doctor or pharmacist if you are taking or have recently taken any other medicines, including herbal products or drugs obtained without a prescription.

Especially, tell your doctor or pharmacist if you are taking:

When administered simultaneously pentoxifylline and antihypertensive, may be required adjusting the dose individually; therefore, must inform your doctor if are taking any blood pressure medicine.

Treatment with pentoxifylline in patients treated with diabetes medications (insulin or oral agents) can enhance the effect of these, so you must inform the doctor if are taking medicine for diabetes.

Similarly, simultaneous treatment of pentoxifylline and theophylline may cause a rise in theophylline levels and thereby increase the side effects of it, so also will need to tell your doctor if you are taking any medication containing theophylline.

Use with food and beverages

This medicine must swallow whole with some liquid (about half a glass of water) during or shortly after a meal.

Special physiological situations

Pregnancy and lactation

In case of pregnancy you should not use this medicine.

If you are lactating, should take into consideration that pentoxifylline passes into breast milk in small amounts. Consult your doctor before taking this drug to assessing the appropriateness of continuing or not treatment.

Children

Do not administer this drug to children.

Driving and using machines

It has not been reported any negative effect of pentoxifylline on the ability to drive car or operate machinery.

How to use pentoxifylline     

Follow exactly instructions of use of pentoxifylline provided by your doctor. Consult your doctor or pharmacist when in doubt. Follow the instructions of the physician in preference to those given in this website, which may be different. It is important to tell your doctor about any illness you suffer, as it could be useful to determine the correct dosage for you.

Your doctor will tell you how long your treatment with pentoxifylline. Do not stop treatment early because you would not get the desired therapeutic effect.

Do not take more medicine than your doctor has recommended.

Swallow the tablet whole with some liquid during or shortly after meals.

Oral dosage for adults and elderly patients:

One tablet of pentoxifylline two or three times a day, depending on the prescribed dosage form , which means a daily maximum two 600-mg tablets or three 400-mg tablets (1200 mg pentoxifylline).

Follow your doctor's instructions regarding the number of tablets you should take and how often, since the dose is decided according to the nature and severity of the disease and the tolerance of each patient.

Dosage for intravenous administration:

It is recommended to administer an infusion of 100-600 mg pentoxifylline once or twice a day. The infusion may be supplemented by oral treatment with 400 mg tablets, resulting the total daily dose 1200 mg pentoxifylline

In advanced cases, particularly in patients with severe pain at rest, gangrene or ulcer, may be administered both a continuous intravenous infusion of up to 1200 mg in 24 hours, as two infusions of 600 mg each during at least 6 hours.

Should not exceed the maximum dose of 1200 mg of pentoxifylline for 24 hours.

The infusion time must be at least 60 minutes for each 100 mg pentoxifilina. It is important you tell your doctor about any illness you suffer, since it could be useful to determine the correct dosage for you.

If you have a severe kidney or liver disease, your doctor will reduce the dose, adjusting it depending on the disease and your drug tolerance.

If you have low blood pressure or are prone to it, or have a disease for which a decrease in blood pressure could be harmful, such as severe coronary heart disease or stenosis, your doctor will prescribe a low initial dose that will increase gradually, according to your improvement degree.

If you think the effect of the medicine is too strong or too weak, consult your doctor.

While you take pentoxifylline     

If a doctor prescribes another medicine, tell you are taking pentoxifylline.

If you feel unwell during treatment with pentoxifylline, immediately consult your doctor.

If you take more than you should:

If you accidentally take more pentoxifylline than you should, may experience nausea, dizziness, palpitations or a drop in blood pressure. Also can occur fever, tremors, hot flashes, loss of reflexes, seizures, skin rashes, vomiting that looks like coffee grounds and unconsciousness.

If you experience these symptoms, consult your doctor or pharmacist immediately or call the National Poisons Information Service in your country. If you have taken a pentoxifylline overdose, go to the emergency department of the nearest hospital. Take the container with you, even if it is empty

If you forget to take pentoxifylline

Do not take a double dose to make up for a forgotten dose.

Side Effects of pentoxifylline     

Like all medicines, pentoxifylline can cause side effects, although not everybody gets them.

The frequency of side effects is classified into:

Very common: affect 1 out of 1-10 patients
Common: affect 1 out of 10-100 patients
Uncommon: affect 1 out of 100-1.000 patients
Rare: affect 1 out of 1.000-10.000 patients
Very Rare: affect less than 1 in 10,000 patients
Incidence not known: Can not be established the true incidence from the available data

When taking high doses pentoxifylline,occasionally may occur:

- Hot flushes,
- Gastrointestinal disorders such as stomach pressure, bloating, nausea, vomiting or diarrhea,
- Heart rhythm disturbances (eg .: palpitations).
