Indications and usage     

Pregabalin belongs to a group of medicines used to treat epilepsy, neuropathic pain and generalized anxiety disorder (GAD).

It is not established the safety and effectiveness of pregabalin in patients under 18 years with peripheral and central neuropathic pain, epilepsy and generalized anxiety disorder.

Peripheral and central neuropathic pain:

Pregabalin is used to treat chronic pain caused by nerve damage. There are several diseases that can cause peripheral neuropathic pain, such as diabetes or shingles. This pain could be described as hot, burning, throbbing pain, shooting pain, pricking, sharp pain, cramping, aching, tingling, numbness and stinging. The peripheral and central neuropathic pain may also be associated with mood changes, sleep disturbance, fatigue, and can have an impact on physical and social functioning and quality of life of patients.

Epilepsy:

Pregabalin is used to treat certain kinds of epilepsy (partial seizures with or without secondary generalization) in adults. Your doctor will prescribe pregabalin to treat epilepsy when current treatment is not controlling properly your condition. You should take pregabalin added to current treatment. This medicine should not be administered alone; it should always be used in combination with other antiepileptic treatments.

Generalized anxiety disorder (GAD):

Pregabalin is used in the treatment of generalized anxiety disorder (GAD). Symptoms of GAD include anxiety and worries which are prolonged and excessive and difficult to control. GAD can also cause restlessness or feeling excitement or nervousness, feeling easily fatigued, having difficulty concentrating, to have mind blank, irritability, muscle tension or sleep disturbance. This is too different from stress and strains of everyday life.

Other ingredients of available commercial presentations:

Each capsule contains 25 mg, 50 mg, 75 mg, 100 mg, 150 mg, 200 mg, 225 mg or 300 mg pregabalin. The other ingredients are lactose monohydrate, maize starch, talc, gelatin, titanium dioxide (E171), sodium lauryl sulfate, anhydrous colloidal silica, black printing ink (containing shellac, black iron oxide (E172), propylene glycol, potassium hydroxide) and water.

Important information     

This medicine contains lactose. If your doctor has told you have an intolerance to sugars, contact him before taking it.

Before taking the drug     

Do not use pregabalin

If you are allergic (hypersensitive) to pregabalin or any of the other ingredients of the medicinal product prescribed to you. If you think may be allergic, ask your doctor.

Allergy symptoms may include:

- Asthma attacks with shortness of breath, audible wheezing or rapid breathing.
- More or less sudden swelling of face, lips, tongue or elsewhere in the body. It is especially critical if it affects the vocal cords.
- Hives, itching, rash
- Anaphylactic shock (loss of consciousness, paleness, sweating, etc.).

Take special care with pregabalin

Some patients treated with pregabalin have reported symptoms suggesting an allergic reaction. These symptoms include swelling of the face, lips, tongue and throat, as well as diffuse skin rashes. If you experience any of these symptoms should seek immediate medical assistance.

Pregabalin has been associated with dizziness and somnolence, which could increase the occurrence of accidental injury (falls) in elderly patients. Therefore, you should exercise caution until be familiar with the effects that can cause the drug.

Pregabalin may be associated with blurred vision and other vision changes. If you experience any change in vision, you should inform your doctor immediately.

Diabetic patients who gain weight while are taking pregabalin may need a change in their diabetes medicines.

Certain side effects, such as drowsiness, may be more frequent because patients with spinal cord injury may be taking other medications to treat their symptoms, for example, pain or spasticity (muscular stiffness), with similar side effects to those of pregabalin, increasing severity when taken together.

There have been reports of congestive heart failure in some patients treated with this drug. Most of them were elderly with cardiovascular conditions. Before starting treatment with this medicine, should tell your doctor if you have a history of heart disease.

Remember that your doctor has prescribed this medicine only for you. Never give it to someone else.

Taking other medicines

Tell your doctor or pharmacist if you are taking or have recently taken any other medicines, including herbal products or drugs obtained without a prescription.

Especially, tell your doctor:

Pregnancy and lactation

Always consult your doctor or pharmacist before taking any medicine during pregnancy or lactation.

Pregabalin should not be taken during pregnancy, unless your doctor has told you. Women of childbearing age must use an effective contraception method.

See your doctor immediately if you become pregnant, you think you might be pregnant or planning to become pregnant while are taking pregabalin.

It is not recommended to breastfeed while taking pregabalin. It is not known whether this drug is excreted in breast milk.

Use in children

It is not established safety and efficacy of pregabalin in children and adolescents (under 18). Therefore, pregabalin should not be used in this age group.

Driving and using machines

Pregabalin can cause dizziness, drowsiness and impaired concentration. You should not drive, operate heavy machinery or engage in other potentially dangerous activities until you know whether this medication affects your ability to perform these activities.

How to use pregabalin     

Follow exactly instructions of use of pregabalin provided by your doctor. Consult your doctor or pharmacist when in doubt. Follow the instructions of the physician in preference to those given in this website, which may be different. Your doctor will determine what dose is appropriate for you. This medicine is for oral use.

Peripheral and central neuropathic pain, epilepsy and generalized anxiety disorder:

Take the number of capsules as instructed by your doctor. The dose, which has been adjusted for you and your condition, will generally be between 150 mg and 600 mg daily.

Your doctor will indicate you taking pregabalin two or three times a day. If you should take it twice a day, take it once in the morning and once in night, at about the same time every day. For three times a day, take it at morning, noon and evening, at about the same time each day.

If you are an elderly patient (over 65 years), you should take pregabalin normally, except if you have kidney problems.

Your doctor may prescribe other dosing regimen or different dose if you have kidney problems.

Swallow the capsule whole, with a little water.

Continue to take pregabalin until your doctor tells you to stop.

If you think the effect of the medicine is too strong or too weak, consult your doctor.

While you take pregabalin     

If a doctor prescribes another medicine, tell you are taking pregabalin.

If you feel unwell during treatment with pregabalin, immediately consult your doctor.

If you take more than you should:

Consult your doctor or pharmacist immediately or call the National Poisons Information Service in your country. If you have taken an overdose of pregabalin, go to the emergency department of the nearest hospital. Take the container with you, even if it is empty.

If you forget to take pregabalin

It is important to take pregabalin regularly, at the same time each day. If you miss a dose, take it as soon as you remember unless it is near the time of the next dose. In that case, continue with the next dose as normal.

Do not take a double dose to make up for a forgotten dose.

If you stop taking pregabalin

Do not stop taking this medicine unless your doctor tells it. If you stop treatment, this should be done gradually over a minimum of one week.

Upon completion of pregabalin treatment both long and short term, you should be aware that may experience certain side effects. These include sleep problems, headache, nausea, diarrhea, flu-like symptoms, nervousness, depression, pain, sweating and dizziness.

Until now, It is unclear whether these symptoms occur more commonly or are more severe if you have been taking pregabalin for a long period of time.

Side Effects of pregabalin     

Like all medicines, pregabalin can cause side effects, although not everybody gets them.

- Dizziness, tiredness

Common side effects (affect 1 in 10-100 patients):

- Increased appetite
- Euphoria, confusion, changes in sexual appetite, irritability
- Disturbance in attention, clumsiness, memory impairment, tremors, slurred speech, tingling
- Blurred vision, double vision
- Vertigo
- Dry mouth, constipation, vomiting, flatulence
- Difficulty in erection (erectile dysfunction)
- Swelling of the extremities, feeling drunk, abnormal gait
- Weight gain

Uncommon side effects (affect 1 in 100-1000 patients):

- Loss of appetite
- Change in self-perception, restlessness, depression, agitation, mood changes, insomnia aggravation, difficulty finding words, hallucinations, abnormal dreams, panic attacks, apathy
- Difficulty thinking, numbness, unusual eye movement, jerky movements, reduced reflexes, hyperactivity, dizziness on standing, sensitive skin, loss of taste, burning sensation, tremor to move, decreased consciousness, fainting
- Dry eyes, swollen eyes, sore eyes, tired eyes, or runny eyes
- Increased heart rate
- Flushing, hot flushes
- Difficulty breathing, nasal dryness
- Swollen abdomen, increased saliva production, heartburn, numbness around the mouth
- Sweating, rash
- Muscle spasms, joint swelling, muscle cramps, muscle pain, joint pain, back pain, pain in limb, muscle stiffness
- Difficult or painful urination, inability to hold urine,
- Weakness, fall, thirst, chest tightness

Rare side effects (affect 1 in 1.000-10.000 patients):

- Decreased blood pressure, coldness in hands and feet, increased blood pressure
- Cough, nasal congestion
- Muscle damage
- Chest pain, cessation of menstruation
- High blood sugar level
- Increased sensitivity to noise
- Heart rhythm disturbances
- Kidney failure, reduced urine output

Have been reported other adverse effects during the post-marketing phase of the drug, including:

- Heart failure,
- Unconsciousness,
- Hypersensitivity and allergic reactions (which may include swollen face, swollen tongue (angioedema), difficulty breathing (asthma, bronchospasm), itching,
- Eye inflammation (keratitis),
- Severe skin reaction characterized by rash, blisters (Stevens-Johnson syndrome), peeling skin and pain,
- mental impairment,
- Urinary retention,
- Diarrhea, headache and nausea.

If you experience swollen face or tongue or if your skin becomes red with blistering or peeling, should seek immediate medical assistance.

Certain side effects, such as drowsiness, may be more frequent because patients with spinal cord injury may be taking other medications to treat, for example, pain or spasticity (muscular stiffness), with similar adverse effects to pregabalin so that the intensity of these effects may be increased when taken together.
