Indications and usage ▲

The active ingredient domperidone belongs to a group of medicines called dopamine agonists, acting as a gastric motility agent.

Domperidone is indicated for the relief of certain gastrointestinal symptoms such as nausea, vomiting, fullness and bloating stomach, abdominal discomfort, and regurgitation of stomach contents.

Other ingredients of available commercial presentations:

Tablets:

Each tablet contains 10 mg of domperidone. The other ingredients are lactose monohydrate, corn starch without gluten, microcrystalline cellulose (E460), pregelatinized potato starch, povidone, magnesium stearate (E572), sodium lauryl sulfate, and hydrogenated castor oil. Coating: Sodium lauryl sulfate, methylcellulose.

Oral solution 1 mg/ml:

Besides the active ingredient, the solution contains carbomer, sodium saccharin (E 954), microcrystalline cellulose (E 460), sodium carboxymethylcellulose (E 466), sorbitol (E 420), sodium methyl para-hydroxybenzoate (E 219), sodium propyl para-hydroxybenzoate (E 217), polysorbate 20 (E 432), strawberry essence and water.

Capsules:

This dosage form contains 10 mg domperidone. The other ingredients are lactose, magnesium stearate, talc, gelatin, titanium dioxide, erythrosine, red iron oxide and black iron oxide.

Suppositories:

Each suppository contains 30 or 60 mg of domperidone. The other ingredients are: semisynthetic glyceride, sorbitan monooleate, tartaric acid, butylated hydroxyanisole (BHA), polyethylene glycol 1000 and 400.

Important information ▲

Oral solution 1 mg/ml:

This dosage form contains sorbitol and may be unsuitable for people with fructose intolerance. It also contains methyl para-hydroxybenzoate and propyl para-hydroxybenzoate which may cause allergic reactions (possibly delayed). Also may produce side effects as headache, stomach upset and diarrhea.

Tablets and capsules:

This medicine contains lactose. If your doctor has told you that you have an intolerance to some sugars, contact your doctor before taking this medicine.

Suppositories:

This dosage form contains butylated hydroxyanisole excipient, which can cause local skin reactions (eg contact dermatitis or irritation of eyes and mucous membranes).

Do not use domperidone

If you are allergic (hypersensitive) to the active substance domperidone or any of the other ingredients of the medicinal product prescribed to you. If you think you may be allergic, ask your doctor.

Allergy symptoms may include:

- Asthma attacks with shortness of breath, audible wheezing or rapid breathing.
- More or less sudden swelling of the face, lips, tongue or elsewhere in the body. It is especially critical if it affects the vocal cords.
- Hives, itching, rash
- Anaphylactic shock (loss of consciousness, paleness, sweating, etc.).

Nor take this medicine if:

- You suffer a disease of the pituitary gland called prolactinoma.
- You have severe abdominal pain or blood in stool (black stools) or vomiting blood.

Take special care with domperidone

If you have kidney problems, you should consult with the doctor before using domperidone. If you are taking domperidone long time, your doctor may consider necessary a dose adjustment.

Patients with liver problems should not take domperidone.

Lactation

May appear small amounts of domperidone in breast milk, so that, if you are breastfeeding, it is not recommended taking this drug.

Driving and using machines

At normal doses, domperidone does not affect the ability to drive or use machines. If you notice symptoms such as drowsiness, disorientation or lack of coordination, do not drive or use machines. You should try to not perform tasks which may require special attention (driving or operating machinery) until you know how you tolerate the drug.

How to use domperidone ▲

Adults and adolescents (over 12 years weighing 35 kg or more)

The drug is taken orally. The usual dose is 10 or 20 mg (one or two tablets), 3 or 4 times daily. The maximum daily dose is 80 mg (two tablets, 4 times a day). This medicine is most effective if is taken a quarter of an hour or half an hour before meals and, if necessary, before bedtime.

When using suppositories, the dose is 60 mg (adults) every twelve hours.

Patients with kidney disease

If you have a kidney disease, your doctor may recommend that you take a lower dose or take the medication less frequently.

Infants and children

0.25 to 0.5 mg / kg three or four times a day with a maximum daily dose of 2.4 mg / kg (not to exceed 80 mg per day).

Infant suppositories: 30 mg/unit are not suitable for use in children weighing less than 15 kg. Children over 15 kg can receive 1 suppository of 30 mg twice daily.

If you have taken more domperidone than you should, you may experience drowsiness, confusion, uncontrolled movements, such as irregular eye movements or postural changes (twisted neck).

In the case of overdose or accidental ingestion consult the Poison Control Center in your country.

If you forget a dose:

If you forget a dose, take the medicine as soon as possible, continuing the treatment prescribed as usual. However, if you are close to the next dose, it is best not to take the missed dose and take the next at the time planned.

Do not take a double dose to make up for missed doses.

Side Effects of domperidone ▲

Like all medicines, domperidone can cause side effects, although not everybody gets them.

The frequency of side effects is classified into:

Very common: affect 1 out of 1-10 patients
Common: affect 1 out of 10-100 patients
Uncommon: affect 1 out of 100-1.000 patients
Rare: affect 1 out of 1.000-10.000 patients
Very Rare: affect less than 1 in 10,000 patients
Incidence not known: Can not be established the true incidence from the available data

Rarely may occur intestinal discomfort . This discomfort usually disappears alone within a short period of time. Also, may appear breast enlargement (gynecomastia) or secretion of milk from the nipple (galactorrhea) and, In women, the menstrual period; may occasionally be irregular or not be present (amenorrhea).

There have been reports of decreased libido. If you believe are problematic, ask your doctor or pharmacist.

Very rarely can produce strong intestinal cramps, Uncontrolled, movements such as irregular eye movements, postural changes with a twisted neck (tardive dyskinesia), tremors and stiff muscles, which disappear when the treatment with domperidone is stopped. It can also occur very rarely itching or rash, and choppy or noisy breathing and/or facial swelling (angioedema).
